IDF21-0279 Association between Renal Tubulointerstitial Inflammatory Cells Infiltration and Renal Outcome in Diabetic Nephropathy

چکیده
Background: Diabetic nephropathy (DN) continues to be the leading cause of end-stage renal disease (ESRD) worldwide, and increased amount evidence supports that inflammation plays an important role in DN progression. Aim: Therefore, we aimed explore relationships between tubulointerstitial inflammatory cells infiltration kidney prognosis patients. Method: A total 384 Chinese participants with type 2 diabetes (T2D) biopsy-confirmed January 2008 July 2019 were enrolled. Cox proportional-hazards regression analysis was used for analyzing association outcome including plasma cells, lymphocytes, monocytes, neutrophils eosinophils. Single-cell RNA sequencing human conducted compare gene expression healthy subjects. Results: Among five kinds only presence found independently associated incident ESRD patients after adjusting model 1 (age, sex, baseline 24-hour proteinuria, estimated glomerular filtration rate, glycated hemoglobin, triglyceride, hazard ratio (HR)=1.44, p=0.037) (parameters plus complement3 Deposit+ glomeruli, class, interstitial inflammation, fibrosis tubular atrophy (IFTA) as well arteriolar hyalinosis, HR=1.44, p=0.047). Moreover, percentages samples by single-cell higher compared control. RPS26, HLA-DRB5, HLA-DQA2, MT-CO2, MT-CO3, CD52, LINC01781, SH3BGRL3, C8orf59, MZT2B up regulated cell group SLC20A1, RGS2, MIDN, TLE3, HLA-DRB1, SKIL, MZB1, ZFP36, HSPA1B HERPUD1 down group. Discussion: The independent prognostic risk factor future T2D DN.

Renal transplantation in diabetic nephropathy.
Forty diabetics who had developed end-stage renal failure from diabetic nephropathy and underwent renal transplantation have been followed up from one to six years. After one and two years 63% and 42% survived (45% and 33% respectively with functioning kidneys). Older patients, those with coronary and peripheral vascular disease, and those with severe neuropathy are prone to higher postoperativ...
متن کاملTubulointerstitial disease in diabetic nephropathy
Diabetes mellitus is the major cause of end stage renal disease (ESRD). We cannot predict which patient will be affected. ESRD patients suffer an extremely high mortality rate, due to a very high incidence of cardiovascular disease. Several randomized, prospective studies have been conducted to quantify the impact of strict glycemic control on morbidity and mortality, and have consistently demo...
متن کاملPathobiology of renal-specific oxidoreductase/myo-inositol oxygenase in diabetic nephropathy: its implications in tubulointerstitial fibrosis.
Renal-specific oxido-reductase/myoinositol oxygenase (RSOR/MIOX) is expressed in renal tubules. It catabolizes myo-inositol and its expression is increased in diabetic mice and in LLC-PK(1) cells under high-glucose ambience. Aldose reductase (AR) is another aldo-keto reductase that is expressed in renal tubules. It regulates the polyol pathway and plays an important role in glucose metabolism, ...
متن کاملCDK5 promotes renal tubulointerstitial fibrosis in diabetic nephropathy via ERK1/2/PPARγ pathway
Cyclin-dependent kinase 5 (CDK5) has been documented in podocyte injuries in diabetic nephropathy (DN), however its role in renal tubular epithelial cells has not been elucidated. We report here that CDK5 is detrimental and promotes tubulointerstitial fibrosis (TIF) via the extracellular signal-regulated kinase 1/2 (ERK1/2)/peroxisome proliferator-activated receptor gamma (PPRAγ) pathway in DN....
متن کاملAssociation between Oxidative Stress, Systemic Inflammation and Renal Function in Type 2 Diabetic Nephropathy
Diabetic nephropathy (DN) is a metabolic disorder with high rate of morbidity and mortality. Global pattern of diabetic nephropathy incidence is not entirely explored, but its incidence has already occurred widely and is estimated to be increased in prevalence [1]. However, DN is one of the main microvascular diabetic complications that leads to renal failure worldwide [2-5]. While, DN occur in...
